The video, produced by advertising agency Hurrell Moseley Dawson & Grimmer, urges the MySpace community to sign an online petition to fight climate change by addressing rainforest destruction.
Prince Charles announced last week that he is also working on a climate change-themed film and book, called 'Harmony', scheduled for release next year.
MySpace claims it is the first time a member of the Royal family has partnered with a social media platform to promote a personal cause.
In the video, Prince Charles said: "One of the internet's great strengths is that it can enable diverse communities to come together to ensure that everybody's views and actions can really be made to count.
"It provides the potential to create global determination for change on a vitally important issue."
The Prince's embrace of social networking is a diversion from the message he delivered last year, when he said technology dependence had left many people detached from nature.
The MySpace appearance follows in the footsteps of the Queen, who sent her first official email last week to mark the 60th anniversary of the Commonwealth.
