Organised by the London & Surrey Cycling Partnership – a joint venture between the organisers of the London Marathon and The Tour of Britain – the event forms part of a weekend of cycling events taking place in the capital over 3-4 August.
Only 20,000 places are available for the challenge, which is based on the route of the 2012 Games cycle road race and begins at the Queen Elizabeth Olympic Park on 4 August.
The ballot for entries closes later today (4 January), however unsuccessful applicants will still have the opportunity to cycle for one of the 300 charities signed up to participate.
More than 200,000 visitors are expected to flock to London to spectate or take part in the weekend, which also includes an eight-mile family fun ride.
Speaking on behalf of the London and Surrey Cycling Partnership, Hugh Brasher, race director of the Virgin London Marathon, said: "We are ecstatic with the response we have had for the first year of this iconic event. RideLondon-Surrey 100 is destined to do for cycling what the London Marathon did for running in the 80s and take it to a new level. People have been inspired by the thought of riding the route made famous by the London 2012 games and have been signing up in record numbers."
