The Brighton-based agency, which specialises in search marketing, will be opening premises in London's Victoria, and said it would be expanding its senior level management to handle its global business.
The office will also serve as the headquarters for US digital marketing firm iCrossing, which was acquired by Spannerworks for more than £10m in February.
Spannerworks said it had been reorganising its departments internally to ensure all of its clients had access to integrated search teams, and opening a London office was part of that move.
Arjo Ghosh, chief executive officer at Spannerworks, said: "We already have a well-established reputation in London, but the new office will enable us to respond to requests for integrated global campaigns far more effectively."
The agency, which was founded in 1997, lists BT, HBOS and Best Western Hotels among its clients.
Last week, Spannerworks and iCrossing created its first search optimisation campaign for online travel firm STA Travel.
