Headlining the festival on Sunday 14 July at the Queen Elizabeth Park in east London, Timberlake and Jay Z will together perform a special preview of their North American stadium tour Legends of Summer.
The two artists were already confirmed to appear at the festival independently, with Timberlake headlining on Friday 12 July, and Jay Z on Saturday 13 July.
According to the festival’s organisers Live Nation, there has been phenomenal demand for tickets, with record sales for the first two days.
Acts also confirmed for the third day include A Tribe Called Quest, in its first UK performance in 20 years, as well as Nas, A$AP Rocky, and Rizzle Kicks.
The park is managed by London Legacy Development Corporation and chief executive Dennis Hone said: "It’s a credit to Queen Elizabeth Olympic Park that this year’s Yahoo! Wireless Festival is the fastest selling in the event’s history and we are delighted that both Justin Timberlake and Jay Z, will be performing a third date here in July with a preview of their Legends of the Summer Tour."
The festival is moving to the Queen Elizabeth Park – formerly the Olympic Park – this year from Hyde Park after AEG Live won the contract to run a limited series of concerts in the central London park.
Additional acts confirmed to join Justin Timberlake for his solo headline show on 12 July are Ke$ha, Wretch 32, Conor Maynard, Bingo Players, and Iggy Azalea.
The bill also features Snoop Dogg, Trey Songz, John Legend, Miguel and special guest Frank Ocean.
Meanwhile, Macklemore and Ryan Lewis, Flux Pavilion and Zedd join already announced Emeli Sande, Rita Ora, Kendrick Lamar, Miguel, DJ Fresh/Live, and Calvin Harris on 13 July.
