The singer and sometimes-actress joins the ranks of unlikely celebrities to promote Nintendo's handheld console, including actresses Nicole Kidman, Lisa Kudrow and Liv Tyler.
Details of the ad itself, which launches on March 30, are thin on the ground. However, the spot will show Beyonce playing the game with her real-life nephew Juelz and the internet is rife with links to footage from the ad shoot.
The video shows Beyonce on location in a New York apartment with views across the Manhattan skyline. She is filmed as she is made up for the shoot, playing 'Rhythm Heaven', autographing a number of handheld consoles and playing the game with Juelz.
In the game, players must tap the console's stylus on the second screen in time to quirky musical and visual prompts on the top screen.
The film also includes an interview with Beyonce in which she talks about her memories of playing computer games when she was a girl and praises 'Rhythm Heaven'.
In a recent interview with American magazine Entertainment Weekly, Beyonce admitted to being something of a gamer. She said: "I like all the Wii games. Love 'Guitar Hero'. ['Rhythm Heaven'] reminds me a bit of 'Guitar Hero', which is great, because I don't want to carry around a big guitar.
"Growing up, I liked Tetris. I even like BrickBreaker on the BlackBerry."
'Rhythm Heaven' has sold 1.6m copies in Japan since its launch in July 2008.
