To showcase the sport, NFL UK will be hosting a 'Block Party' to celebrate the following day’s game between the Minnesota Vikings and Pittsburgh Steelers.
The free event, taking place between 12 noon and 6pm, will feature music, food, cheerleaders, team drum-lines and interactive games, along with other popular elements of American football.
Players from the Vikings and Steelers will be taking time out from their preparations for the following day’s showdown at Wembley Stadium to celebrate with fans on Regent Street.
Alistair Kirkwood, managing director of NFL UK, said: "We are really looking forward to giving visitors to Regent Street the opportunity to sample the excitement and razzmatazz of the NFL.
"The range of activities planned will offer something for everyone; whether a die-hard NFL fan or those who are simply looking for a fun day out or are curious to find out a little bit more about the sport."
Regent Street will remain a traffic-free area from 4am until midnight on the day of the event.
Previous events which have resulted in Regent Street becoming traffic free included Summer Streets in July, in partnership with the Mayor of London, which provided visitors and shoppers with a range of activities over four consecutive weekends.
Future traffic-free events planned include the annual Regent Street Motor Show on 2 November, and the Regent Street Christmas Lights switch-on taking place on 9 November.
