It is first time that the IPC Media music title has launched a special standalone title for the UK's top music festival.
The NME, which celebrated its 50th birthday last year, is promising never-before-seen backstage photos of artists and revellers out in the fields, as well as articles on bands set to play this year.
Steve Sutherland, editorial director of the NME, said: "The NME has celebrated every Glastonbury since 1970 and our photo archive of the festival is the world's best.
"We have all the tall tales, all the highs and all the disasters on record and we are pleased to publish this magazine, which will interest seasoned festival veterans, this year's Glasto virgins and innocent bystanders alike."
Rival publisher Emap has also got in on the act and, for the second year running, is to publish a daily newspaper version of its music monthly Q at this year's event and a 68-page review of the festival.
Emap's ticket website Aloud.com was the official online retailer of Glastonbury tickets this year.
Tickets for this year's festival, to be held on June 27-29, sold out in 24 hours.
The line-up has not yet been confirmed but it is expected to include Radiohead, REM, Moby, Turin Brakes, Coldplay, David Gray, Suede and the Polyphonic Spree.
The magazine goes on sale June 2.
