The UTV-owned station was awarded the contract in October last year. it won one of seven 32-game Premier League packages for three seasons from 2007/2008.
The other six packages went to the BBC, which has been the only national radio broadcaster to cover live, top-flight games since it first aired live commentary on Arsenal vs Sheffield United in 1927.
The station plans to fit pre-match build-up and post-match phone-ins around the live games.
In September, TalkSport won the national radio commentary rights for Chelsea, Arsenal and Liverpool UEFA Champions League matches. The station has also netted London-only coverage of all Chelsea and Arsenal Premiership home games
MindShare is currently defending its position on the £50m pan-European media planning and buying account, due to a pitch announced earlier this month.
TalkSport confirmed that the media planning and buying brief for promoting the Premiership broadcasts had been awarded to MindShare.
