The show has had a change of format for 2009, which will see contestants perform live in front of judges Simon Cowell, Cheryl Cole, Dannii Minogue and Louis Walsh and a 1,300 strong audience.
From the 3-5 July the venue's Exchange Hall will be transformed into The X Factor's set.
It is hoped the live auditions will give the contestants an added boost with public support and will help to improve performances.
"Music is a significant part of the venue's past and with the The X Factor live auditions and more concerts planned for December it is set to be a part of our future as well," said Manchester Central chief executive Lesley Tomlinson said. "It is very exciting for us and the whole team is buzzing with excitement!"
