The partnership with John Smith's Grand National will run between 2010 and 2012.
The tie-up will include both newspaper and online reader offers, ticket promotions, competitions and editorial support.
The Daily Mirror will have a presence at the Liverpool course through advertising hoardings.
It will also have the opportunity to distribute the paper to Aintree race-goers.
The move marks a ramping up of its existing relationship with the Grand National.
It has previously teamed up with The John Smith's People's Race – an amateur charitable flat race for the public during the event.
Richard Wallace, editor of the Daily Mirror, said: "This is an ideal partnership for our readers and is further evidence of the Mirror's commitment and long-term investment in racing."
The Grand National will take place from 8 April and 10 April.
