The service, called City PM, launches this week, and will summarise the day's City news as well as previewing the next day's paper. It will be available for free download every weekday from 5pm.
Research carried out by City AM revealed that more than half of its readership owns an iPod or MP3 player.
The podcast is being launched in association with retail investment fund management group Threadneedle Investments, which will have a sponsored message at the start and finish of the five-minute broadcasts.
Presented in the style of a radio news bulletin, the podcasts will be read by financial journalist Giles McMullen.
The downloads will be available from the websites of both City AM and Threadneedle Investment.
City AM hopes the service will help it reach members of London's business community outside its City of London and Canary Wharf distribution points.
The paper, which was launched in September 2005, is distributed by hand on weekdays between 6.30am and 10am in the City and business lounges at London's airports. It claims to have a total daily distribution of about 70,000, with a readership of nearly 100,000.
