The contract stipulates that Associated Newspapers can no longer distribute copies of its freesheet via its Evening Standard vendors.
Ian Clark, general manager of thelondonpaper, said: "The potential readers in Canary Wharf are an advertiser's dream."
As well as the Evening Standard, thelondonpaper will join City AM, which distributes more than 10,000 copies each morning.
This is the first time News International's paper has been available on the private estate, which is owned by Canary Wharf Group and has a footfall of more than 650,000 shoppers each week.
In addition, more than 90,000 people are employed on the 97-acre estate, many of whom leave their offices and go directly to the underground without crossing public land, where the freesheets are currently distributed.
News International will initially employ hand distributors to raise the paper's profile inside Canary Wharf, though the paper will largely be distributed in branded bins. It will be available from midday each weekday.
The paper's total distribution will remain at 500,000.
