The fans want other television networks to take over the rights to the show, which has just begun its fourth and final series in the US.
Under the headline "Save Star Trek", the full-page ad calls on fans to sign a petition campaign and attend a rally.
It reads: "Captain Archer and the crew of the NX-01 need your help to continue their journeys..." and tells viewers to tune into the fourth series to boost its ratings. It it hoped the Sci Fi Channel can be persuaded to pick up the show.
The series not been as successful in the ratings as 'Star Trek: The Next Generation', 'Star Trek Deep Space Nine' and 'Star Trek Voyager'. It is the only spin-off to be cancelled before the end of its run.
The show is a prequel to the original 1966-69 'Star Trek', focusing on man's first voyages out into the galaxy.
A massive petition is already under way and fans are organising a rally on February 25 to help save the show, which stars Scott Bakula as Enterprise Captain Jonathan Archer and Jolene Blalock as Vulcan T'Pol.
The show, which averages fewer than 3m viewers a week, is due to come off the air in May. It will begin its UK run on BSkyB later this year.
While the ad might be a long shot, fan power has been successful in the past. When sci-fi series 'Farscape' was cancelled in 2002 on a major cliffhanger, fans used ads and petitions to bring back the show back. 'Farscape' returned last year for a four-hour mini-series.
'Battlestar Galatica' also returned last year backed by fan power. It gained rave reviews for its dark and gritty approach, which was far removed from its camp 70s predecessor.
