The Fifa president said the English league's plans to expand into five foreign territories by playing 10 39th round league games from January 2011, would severely undermine England's chances of hosting the 2018 World Cup.
He described the idea as "an abuse of power" and "unacceptable" and branded the proposals "a joke at the end of carnival season".
Blatter added that the plans, which were unveiled last week by the chief executive of the Premier League, Richard Scudamore, would "never happen" as long as he was president of Fifa, adding that the proposals to expand England's top division "only had business and money" as motivations.
Under Scudamore's proposals, an additional round of 10 English Premier League games would be played across five foreign cities from January as part of an attempt to expand the league's brand presence globally. Scudamore described the plan, which could generate each club involved an additional £5m in revenue, as "an idea whose time has come".
Blatter's criticism follows a week of criticism towards Scudamore's plans from heads of foreign football associations, including Mohamed bin Hammam, president of the Asian Football Confederation, and Michel Platini, president of Uefa, who branded the idea comical.
Under current international football law, all domestic league associations wishing to play competitive games abroad need to have the backing of their counterparts in the chosen country. Blatter said he would advise all domestic governing bodies not to accept the English Premier League's proposals.
In an interview with the BBC, Blatter said: "If you are the most prosperous league in the world, you accept that everyone in football has a responsibility to maintain it not only as a business, but as a game. Then all of a sudden you come out with a project which only has business and money behind it.
"If you go against the authority of Fifa, and the decision-making body which is the executive committee, then you cannot expect them to be in your favour later on. England will not be the only World Cup bid [for 2018]. I cannot imagine that when you go against the deciding body for the decision on the World Cup in an initiative that will not be accepted by this body,that you will enhance your chances."
He added that his first reaction to Scudamore's plans were that it "was a joke at the end of carnival season", adding, "football cannot be like the Harlem Globetrotters or a circus. You must keep the national identity of the clubs."
A spokesman for the English Premier League responded that Scudamore's proposals were "always the starting point in relation to this issue of sanction of our proposed international round... and we will be making no further comment until after that time."
